In the work described on this paper, a highly specific and sensitive enzyme-linked immunosorbent assay (ELISA) for the determination of glycyrrhizin (GL) was developed. To demonstrate the potential of the ELISA, a preliminary pharmacokinetic study of GL in rats and quantitative analysis of GL in several Chinese prescriptions were performed and compared with high-performance liquid chromatography (HPLC). The experimental data indicated that ELISA method exhibits more advantages over HPLC, such as lower detection limit, high specificity, low background and no requirement of sample pre-treatment, and more suitable for the determination of natural components in Chinese traditional medicines.

Two Tube immunoassays have been developed for the detection and quantification of fungal antigens in grape juice. One for Botrytis, the main cause of Bunch rot in grapes in California, and the other for Botrytis plus rot caused by species of Aspergillus and Penicillium, fungi that can also be involved in Bunch rot. The assays, which are rapid, user friendly and do not require laboratory facilities were designed and tested for on-site use at wineries during harvest. Both assays employ highly specific monoclonal antibodies, BC12.CA4, for Botrytis and an hitherto unreported antibody, AF-CA2, for all species of Aspergillus and Penicillium. The protocol for both assays is the same and mirrors that used in plate-trapped antigen (PTA) ELISAs. The antibodies detect water-soluble fungal antigens present in juice from infected grapes that bind rapidly to the walls of Nunc immunosorbent tubes. Saline extracts from freeze-dried mycelial cultures of Botrytis cinerea and Aspergillus niger are used as standards. The assays are very sensitive detecting between 1 and 3% levels of rot in grapes corresponding to 10-30@mg/ml extracts from freeze-dried mycelium. Absorbance values from the Tube assays parallel but do not equal those from PTA-ELISA tests. No problems were encountered with tests carried out on the testing stands at wineries at harvest time by personnel with no scientific background.

An ultra sensitive automated flow fluorescent immunoassay was developed using the KinExA(TM) 3000 system for quantitative analysis of the neonicotinoid insecticide thiamethoxam. Five new monoclonal antibodies were obtained and screened with a competitive ELISA. One monoclonal antibody designated as E6VI was evaluated for sensitivity, selectivity and solvent tolerance with the KinExA. Sensitivity determined from the concentration of half-maximal inhibition (IC”5″0) was obtained by plotting KinExA signals to a four-parameter sigmoidal curve as a function of analyte concentrations. For the most sensitive clone, the IC”5″0 and the limit of detection were approximately 30pgml^-^1 and 16pgml^-^1, respectively. Cross-reactivity was estimated by measuring the equilibrium constants (K”d) for four other neonicotinoid insecticides (clothianidin, imidacloprid, dinotefuran, and acetamiprid). E6VI was very specific to thiamethoxam with <0.11% cross-reactivity for tested neonicotinoids. An excellent correlation (r^2=0.99) was obtained between spiked and measured concentrations of thiamethoxam in stream and tap water, potato, cucumber, and apple samples.

A new method describing direct attachment of carboxylated haptens on a polystyrene support, using 3-aminopropyltriethoxysilane (3-APTES) as a linker, is reported. The hapten coated polystyrene support showed excellent stability as a function of the buffer pH and reaction time, and was successfully used to demonstrate its application in enzyme-linked immunosorbent assay (ELISA).
